چکیده

Natterin is an aerolysin-like pore-forming toxin responsible for the toxic effects of venom medically significant fish Thalassophryne nattereri. Using a combination pharmacologic and genetic loss-of-function approaches we conduct systematic investigation regulatory mechanisms that control Natterin-induced neutrophilic inflammation in peritonitis model. Our data confirmed capacity to induce strong sustained leading systemic inflammatory lung infiltration revealed overlapping paths its control. We found induced extracellular release mature IL-1? production IL-33 by bronchial epithelial cells. dependence both ST2/IL-33 IL-17A/IL-17RA signaling on local neutrophils migration, as well crucial role IL-1?, caspase-1 caspase-11 inflammation. The triggered was gasdermin-D-dependent inflammasome process, despite cells did not die pyroptosis. Finally, mediated non-canonical NLRP6 NLRC4 adaptors through ASC interaction, independent NLRP3. highlight process dependent activation can be target pharmacological intervention accidents T. nattereri, which does have adequate specific therapy.

Inflammasome Sensor Nlrp1b-Dependent Resistance to Anthrax Is Mediated by Caspase-1, IL-1 Signaling and Neutrophil Recruitment

Bacillus anthracis infects hosts as a spore, germinates, and disseminates in its vegetative form. Production of anthrax lethal and edema toxins following bacterial outgrowth results in host death. Macrophages of inbred mouse strains are either sensitive or resistant to lethal toxin depending on whether they express the lethal toxin responsive or non-responsive alleles of the inflammasome sensor...

Ozone-induced IL-17A and neutrophilic airway inflammation is orchestrated by the caspase-1-IL-1 cascade

Ozone is a common environmental air pollutant leading to respiratory illness. The mechanisms regulating ozone-induced airway inflammation remain poorly understood. We hypothesize that ozone-triggered inflammasome activation and interleukin (IL)-1 production regulate neutrophilic airway inflammation through IL-17A. Pulmonary neutrophilic inflammation was induced by extended (72 h) low-dose (0.7 ...

Caspase-1 activation by NLRP3 inflammasome dampens IL-33-dependent house dust mite-induced allergic lung inflammation.

The cysteine protease caspase-1 (Casp-1) contributes to innate immunity through the assembly of NLRP3, NLRC4, AIM2, and NLRP6 inflammasomes. Here we ask whether caspase-1 activation plays a regulatory role in house dust mite (HDM)-induced experimental allergic airway inflammation. We report enhanced airway inflammation in caspase-1-deficient mice exposed to HDM with a marked eosinophil recruitme...

The IL-1-like cytokine IL-33 is inactivated after maturation by caspase-1.

IL-33 is a chromatin-associated cytokine of the IL-1 family that has recently been linked to many diseases, including asthma, rheumatoid arthritis, atherosclerosis, and cardiovascular diseases. IL-33 signals through the IL-1 receptor-related protein ST2 and drives production of pro-inflammatory and T helper type 2-associated cytokines in mast cells, T helper type 2 lymphocytes, basophils, eosin...

The inflammasome sensor, NLRP3, regulates CNS inflammation and demyelination via caspase-1 and interleukin-18.

Inflammation is increasingly recognized as an important contributor to a host of CNS disorders; however, its regulation in the brain is not well delineated. Nucleotide-binding domain, leucine-rich repeat, pyrin domain containing 3 (NLRP3) is a key component of the inflammasome complex, which also includes ASC (apoptotic speck-containing protein with a card) and procaspase-1. Inflammasome format...
